While the original manor house on the site was constructed in the 17th century using the Karoliner style typical of the era, the current structure stands as a testament to late 19th-century architectural ambition. The old manor was demolished in 1878 to make way for the present castle, which was designed by the renowned Swedish architect Johan Fredrik Åbom. Constructed between 1874 and 1882, the building features a distinctive Neo-Gothic style. The interior of the first floor was notably decorated by Carl Grabow, a celebrated painter and interior designer of the time, adding significant artistic value to the residence.

The historical significance of Boo Castle is deeply intertwined with the Swedish barony Hamilton af Hageby. The estate's lineage traces back to 1692 when it was inherited by Anna Flemming, who later married Baron Hugo Johan Hamilton. In 1723, Hamilton established a fideikommiss, a rare legal structure in Sweden that allows an entire estate to be inherited by a single heir rather than being divided. This arrangement has successfully kept the property within the Hamilton family for generations, making it one of the few remaining fideikommiss estates in the country. This continuity provides a unique glimpse into Swedish aristocratic history and inheritance laws.

In the early 20th century, the castle underwent a major restoration between 1925 and 1927 led by architect Ivar Tengbom. During this period, parts of the building were redesigned to reflect 18th-century aesthetics, and new gardens were created by landscape architect Rudolf Abelin. Today, Boo Castle remains a privately owned property under the stewardship of the Hamilton family.
